The church of the Virgin Mary & Charles the Great at Karlov was founded in 1350, shortly after the emergence of the New Town, it was part of the monastery of the Augustinian Canons, built following the Saxon Chapel of Charles the Great. The church was designed on an octagonal floor plan and it was vaulted with a single arched shape on a stellar ribbed framework. At the beginning of the 18th century

the chapel of the Holy Stairs was added with the underground chapel of the Nativity of the Lord, the author of which is probably Jan Santini-Aichl. The main altar was made in Neo-Gothic style in 1872. The furnishings are mostly Baroque, made by Franz Maximilian Kaňka. Near the church, there is a statue from 1837 by Josef Max and it depicts the founder of the church, Emperor Charles the Fourth.
