Since 1569, the Italian Chapel, as suggested by its name, was the Italian community local church. This community was rather large and consisted of Italian brick masons and traders mainly inhabiting the Lesser Town. A "definite appearance", meaning a chapel built in the Mannerist style, was completed around

1590. The chapel was part of the Jesuit college of the Klementinum. The chapel underwent important reconstruction at the beginning of the 18th century, when a new entrance was opened into today's Karlova Street. The construction modifications were supervised by renowned architect František Maxmilián Kaňka.
